What are the advantages and disadvantages for a player of opting in the 3rd year?
Donald is the only one who suffered disadvantages. All the other opters got contracts.
The advantage is that you come up for a 2nd contract sooner and so get paid your market worth sooner rather than being underpaid for your level of play on a rookie contract.
For the team, the advantage is getting a guy you want to keep on a deal that is at least a bit less expensive than the kind of deal he would get in his free agent year.

I don't see what the advantage is for the player if he has no leverage to get a team to pay him his market worth sooner. What can he do to get it? What's his leverage?
"Leverage" may be an overused and overvalued concept.
As I said, several players have exercised the 3 year option and every single one of them, except for Donald so far, got paid.
Here's a partial list:

Teams giving highly-accomplished first-round picks extremely early extensions haven't exploited their leverage. Cowboys center Travis Frederick, Panthers inside linebacker Luke Kuechly, Cardinals cornerback Patrick Peterson and Texans defensive end J.J. Watt became the highest-paid player (by average year salary) at their respective positions. Each player received All-Pro honors in two of their first three years, just like Donald.
