Jourdan has an absolutely excellent day-after-the-game-review that she calls The Pile and I added the following comment to her article:

The lack of preseason reps bit McVay in the butt against the Bills. Especially the OL, where it's really tough to replicate and build a cohesive unit without playing. Yes injuries are a gamble and Bruss the example, but this is not the first time the Rams OL struggled out of the gate. See 2019.

Plus many were just not ready to play. Tutu and David Long needed preseason snaps. The connection between Stafford and Robinson wasn't there. It's a lot to catch up on before playing the Falcons next.

Will McVay change next year? I get that his focus is on avoiding injuries but clearly the Bills were better prepared than the Rams. This is - arguably - the first time he got bit since he won 5 opening games in a row, but it's also the first "real" opening day opponent, as "None of the Rams’ first opponents in those games finished with more than six victories".
